GB/T15670.17-2017 Standard Overview

This standard specifies the basic principles, methods and requirements for mammalian spermatogonia/spermatocyte chromosome aberration tests, and is applicable to the evaluation of the possibility of test substances causing genetic mutations in germ cells during the pesticide registration process.

Explanation of professional terms and practical application cases

Chromosome structural aberration:refers to the structural changes in the metaphase of cell division observed under a microscope, such as chromosome deletion, exchange and internal exchange.

Gap:refers to the damage length of chromosomes and chromatids that is less than the width of a chromatid, which is the smallest form of incorrect arrangement.

For example, if a significant increase in chromosome aberrations or gaps is observed in the mitotic cells in the high-dose group in the experiment, it can be judged that the test substance has a potential mutagenic effect.

Implementation recommendations

  1. Experimental design:A preliminary test should be conducted before the formal test to determine the appropriate dosage range, especially for test substances that are insoluble in water or oil, which need to be prepared with a suspension.
  2. Control group setting:In addition to the negative control group, it is recommended to set up a positive control group (such as cyclophosphamide and mitomycin C) to verify the effectiveness of the test system.
  3. Data analysis:In statistical analysis, in addition to paying attention to the rate of aberrant cells, the dose-response relationship must also be examined. If there is a significant difference and there is a clear dose-dependence, it can be determined as a positive result.
  4. Record preservation:It is recommended to preserve all test data for a long time and ensure that the original records are traceable.
