Introduction
1. Standard Overview
This standard specifies the quarantine identification method for Maize Chlorotic Mottle Virus (MCMV), and is applicable to plant propagation materials such as seeds and seedlings that may carry the virus. The main detection methods include double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA), reverse transcription polymerase chain reaction (RT-PCR) and real-time fluorescence RT-PCR.
2. Detection Principle
The molecular biological characteristics, serological characteristics and biological characteristics of Maize Chlorotic Mottle Virus are the main basis for the formulation of this standard. By detecting the nucleic acid or antigen of the virus, it can be effectively determined whether the sample carries the virus.
3. Comparison of detection methods
| Detection method | Sensitivity | Scope of application | Operation steps |
|---|---|---|---|
| DAS-ELISA | High sensitivity | Suitable for preliminary screening | 1. Coating antibody2. Sample addition detection3. Enzyme-labeled antibody incubation4. Color reaction |
| RT-PCR | Highly specific | Suitable for amplification detection of viral nucleic acid | 1. RNA extraction2. cDNA synthesis3. PCR amplification4. Electrophoresis analysis |
| Real-time fluorescence RT-PCR | Quantitative detection | Suitable for accurate diagnosis | 1. RNA extraction2. cDNA synthesis3. Real-time amplification monitoring |
4. Implementation suggestions
Case study:In actual applications, DAS-ELISA is recommended as a preliminary screening method, and positive samples are further confirmed by RT-PCR or real-time fluorescence RT-PCR. It is recommended to establish standardized operating procedures and regularly train testing personnel.
