《GB/T 36809-2018可可丛枝病菌检疫鉴定方法》由TC271(全国植物检疫标准化技术委员会)归口,主管部门为国家标准化管理委员会。
Introduction
1. Standard Overview
The National Standard of the People's Republic of China GB/T 36809-2018 specifies the quarantine identification method for the cocoa witches' broom pathogen (Moniliophthora perniciosa). This standard is applicable to the laboratory detection of the cocoa witches' broom pathogen and clarifies the complete process from symptom examination to molecular biological identification.
2. Method Principle
The main basis of this standard includes the symptoms of damage, morphological characteristics and cultural characteristics of the pathogen. Appendix A provides detailed taxonomic status, host range and geographical distribution information, see Table C.1 and Figure B.1.
| Project | Theobroma cacao witches' broom pathogen | Theobroma cacao pod rot pathogen |
|---|---|---|
| Scientific name | Moniliophthora perniciosa | M.roreri |
| Asexual state | Monokaryotic hyphae are swollen, curved and entangled, with a width of 5μm to 20μm | Monokaryotic hyphae, with thick conidia wall, light yellow to brown |
Implementation suggestions section
1. Sample collection and storage:
- Disease branches or fruits should be properly packaged and the source information should be recorded.
- Samples should be stored in a refrigerator at 4°C to prevent strain degradation.
2. Molecular biological identification:
- It is recommended to use the CTAB method to extract DNA to ensure purity and concentration.
- Pay attention to the optimization of primer concentration and reaction conditions during PCR amplification.
3. Data analysis:
- BLAST results need to be combined with phylogenetic trees for comprehensive judgment.
- It is recommended to use MEGA software for evolutionary analysis to ensure the confidence of branches.
