《GB/T 37644-2019化妆品中8-羟基喹啉和硝羟喹啉的测定 高效液相色谱法》由TC257(全国香料香精化妆品标准化技术委员会)归口,TC257SC2(全国香料香精化妆品标准化技术委员会化妆品分会)执行,主管部门为中国轻工业联合会。
Introduction
1. Background and significance of the standard
This standard was proposed by China National Light Industry Council and is under the jurisdiction of the National Technical Committee for Standardization of Flavors, Fragrances and Cosmetics. The drafting units of the standard include Suzhou Shipu Testing Technology Co., Ltd., Yangzhou Perfect Daily Products Co., Ltd. and other enterprises and research institutions. The formulation of this standard aims to meet the testing needs of banned substances in my country's "Technical Specifications for Cosmetic Safety (2015 Edition)", especially to standardize the determination methods of 8-hydroxyquinoline and nitrohydroxyquinoline.
2. Detection Principle
| Steps | Description |
|---|---|
| Sample Treatment | Different pre-treatment methods are used according to the type of cosmetics (powder, wax-based, etc.), including ultrasonic extraction and centrifugal separation. |
| High performance liquid chromatography determination | Use an HPLC instrument equipped with a diode array detector to separate and quantify by gradient elution. |
3. Detailed explanation of experimental steps
Sample treatment:For non-powder cosmetics, weigh 0.1~0.2g of sample into a centrifuge tube and add formic acid-acetonitrile solution for ultrasonic extraction. Wax-based samples need to be melted before processing. Powder cosmetics are extracted with methanol-phosphoric acid aqueous solution.
Determination conditions:The chromatographic column is a C18 column (250mm×4.6mm, 5μm), the mobile phase is gradient elution, the detection wavelength is set to 240nm, and the injection volume is 20μL.
4. Standard working curve and recovery rate
| Material type | Additional concentration range (mg/kg) | Recovery rate (%) | Relative standard deviation (%) |
|---|---|---|---|
| Non-wax-based, non-powder cosmetics | 5.0~50.0 | 80~110 | ≤10 |
| Wax-based and powder cosmetics | 10.0~100.0 | 80~110 | ≤10 |
5. Implementation Suggestions
To ensure the accuracy of the test results, it is recommended that:
- Strictly follow the standard operating procedures for sample processing and instrument calibration.
- Regularly verify the validity of the standard working curve.
- Perform quality control on key reagents and consumables (such as filter membranes and chromatographic columns).
