Introduction

1. Background and purpose of the standard

With the widespread use of silver nanoparticles in daily necessities and medical devices, their potential risks to the human body have received increasing attention. This standard aims to provide a method for accurately measuring the silver content in biological samples, filling the gap in relevant national standards.

2. Core technology: Inductively coupled plasma mass spectrometry (ICP-MS)

ICP-MS is an efficient elemental analysis method suitable for the detection of trace and trace metals. This standard adopts the internal standard method, which effectively suppresses the matrix effect and monitors the instrument signal drift by adding internal standard elements (such as indium and rhodium) online.

3. Experimental process and key steps

Steps Operation content Precautions
Sample pretreatment Microwave digestion, acid removal, and volume determination Pay attention to the complete digestion of the sample and the accuracy of the volume determination
Standard curve drawing Prepare a series of silver standard solutions, and establish a standard curve with concentration as the horizontal axis and signal ratio as the vertical axis The correlation coefficient should be ≥0.99
Sample measurement Add internal standard solution online to measure the signal intensity of the silver element in the sample The blank value of the entire program must meet the requirements

4. Data Analysis and Result Processing

Calculate the total mass and unit mass content of silver according to the formula: m = ρ_d × f × (m₁ / m₂ × 1000), X_i = m / m_T

5. Accuracy and Precision Analysis

The recovery rate of the blank spiked sample should be within the range of ±20% (relaxed to ±25% for low concentrations), and the precision calculation formula is: ε = [√(Σ(m_i - μ)^2 / (n-1))] / μ × 100%

6. Implementation Suggestions

Case Study: Detection of Biological Tissue Containing Silver-Coated Gold Nanorods

The silver content in rat kidney tissue was determined by standard method. The results showed that the silver concentration was 12.5 μg/L and the silver content per unit mass was 10.3 μg/g, which met the precision and accuracy requirements of the method.
