Introduction

Technical Background of the Standard

This standard established for the first time an indicator plant method system based on wheat coleoptile, Amaranthus caudatus cotyledon and barley aleurone layer, solving the problems of poor repeatability and low standardization of traditional bioassay methods. The standard drafting team integrated multidisciplinary technologies such as plant physiology, analytical chemistry and metrology, and established the reproducibility of the method (RSD≤20%) through 5 years of verification.

Key Operation Points

Auxin Determination Case

Use class=instrument>Changmai 6135 variety of wheat, the following must be strictly controlled:
1. Light intensity <5 lux during dark culture
2. Length of coleoptile segments 6±0.2 mm
3. Pre-culture buffer replacement frequency 30 min/time

Buffer preparation specifications

Phosphate-citrate buffer (pH5.0) must be prepared and used immediately, where:
- The ratio of potassium dihydrogen phosphate (1.80 g/L) to citric acid (1.02 g/L) is strictly 1:0.567 (w/w)
- Sucrose concentration of 20 g/L can maintain osmotic pressure balance


Implementation suggestions

  1. Quality control: The recovery rate of the standard sample (85-115%) must be measured simultaneously for each batch of experiments
  2. Equipment calibration: The wavelength accuracy of the UV spectrophotometer needs to be calibrated with potassium dichromate solution every day
  3. Data verification: It is recommended to use Excel LINEST function for regression analysis, and it is effective only when R2≥0.98

Technology evolution analysis

Compared with the traditional mung bean hypocotyl bending method, this standard has the following advantages:
1. The detection sensitivity is increased by 102 times (the minimum detection limit of auxin is 3.16×10-7 mg/mL)
2. The detection cycle is shortened by 50% (the cytokinin determination only takes 72 hours)
3. For the first time, the comparable measurement of the activity values of the three hormones is achieved
