Introduction
Technical background of the standard
As the first special test method for active ingredients in seaweed in my country, this standard fills the technical gap in the synergistic detection of alginate and mannitol. Compared with the original method, the 2018 version adds equipment specifications such as vacuum filtration pump, and the precision requirement is increased to 2%.
Comparison of core detection methods
| Detection items | Principle | Key reagents | End point determination |
|---|---|---|---|
| Alginate | Acid conversion-calcium replacement titration | Calcium acetate solution (0.1mol/L) | Phenolphthalein turns red for 30s |
| Mannitol | Periodic acid oxidation-reduction | Sodium thiosulfate (0.1mol/L) | Starch blue disappears |
Key operating points
Alginate test
The sample needs to be processed with a 60 mesh sieve, and the hydrochloric acid soaking stage needs to be strictly controlled for 12h±0.5h. Actual cases show that the optimal temperature for calcium replacement reaction of laver samples is 25±2℃.
Mannitol test
It is recommended to use a constant temperature water bath to control the temperature difference ≤1℃ when extracting in a boiling water bath. Comparative data from a kelp company showed that the precision of sodium thiosulfate titration is best when the rate is controlled at 1 drop/2 seconds.
Implementation recommendations
- Reagent management: Calcium acetate solution needs to be freshly prepared every month, and periodic acid solution should be stored away from light
- Equipment calibration: The burette should be checked for tightness every month, and the balance should be calibrated daily
- Quality control measures: Each batch of testing should include parallel samples of standard substances
