Introduction

Analysis of the core content of the standard

This standard adopts a high performance liquid chromatograph in conjunction with a fluorescence detector to establish a detection method for malachite green in aquaculture environments (water bodies/sediment), with a quantitative limit of 0.1 μg/L (water bodies) and 1.0 μg/kg (sediment).

Key operating points

Sample processing specifications

Water samples must be filtered through a 0.45 μm filter membrane, and potassium borohydride solution must be added to reduce the latent malachite green; sediment samples must be extracted with a dichloromethane-acetonitrile mixture using an ultrasonic cleaner.

  • Mobile phase: acetonitrile-0.125mol/L ammonium acetate buffer (80:20)
  • Detection wavelength: 265nm (excitation)/360nm (emission)

Standard implementation recommendations

1. The laboratory should regularly verify the recovery rate of the solid phase extraction device, and it is recommended to perform matrix spike experiments for each batch of samples;
2. Note that the standard solution must be prepared and used and stored away from light for no more than 3 months;
3. The water content of the sediment test must be measured simultaneously for result correction.
