Standard Background and Scope of Application
SN/T5135-2019 was issued by the General Administration of Customs to develop a standardized method for quarantine identification of Fruticicola fruticum. This species belongs to the family of Helicidae, class of Gastropoda, class of Mollusca, order of Pulmonaria, and is included in the list of quarantine pests for imported plants in my country.

Typical case: In 2018, live Brown snails were intercepted from imported wood at Dalian Port. The species was confirmed by the number of shell layers and reproductive anatomical characteristics, and the final determination was made after COI gene sequence comparison (similarity ≥ 95%).
Molecular detection technical specifications
The standard stipulates the use of COI gene barcode for molecular identification:
- Primer sequence: LCO1490/HCO2198
- Amplification conditions: 94℃ pre-denaturation for 5min, 30-35 cycles (94℃50s→45℃30s→72℃50s)
- Expected product: 600-700bp fragment
The similarity with the GenBank reference sequence (AY546264.1) must be ≥95% for confirmation.
Quarantine treatment recommendations
- Immediately soak in 75% alcohol for 24-48 hours when living snails are found
- Improve wooden packaging with methyl bromide 48g/m³, 24h
- Establish snail stress resistance monitoring files
