Introduction

Analysis of the core content of the standard

In view of the particularity of African swine fever virus detection, this standard establishes a biosafety protection system from three dimensions: laboratory hierarchical management, sample full process control and disinfection and sterilization specifications:

Virus isolation laboratory Must be carried out in a BSL-3 laboratory Level 3 protection
Nucleic acid testing laboratory Must meet BSL-2 standards Level 2 protection
Serological testing laboratory Need to be equipped with a biological safety cabinet Enhanced Level 2

Key technical points

1. Sample handling specifications

All operations in the biological safety cabinet must comply with the following:

  • Tissue samples must be made into a 10% homogenate and inactivated at 60℃ for 30 minutes
  • Anticoagulated blood must be directly packaged and inactivated, and non-anticoagulated blood must be allowed to stand for 12-24 hours to separate serum
  • Tick samples must be pre-cooled and ground, and centrifuged at 1000g to obtain the supernatant

2. Disinfection and sterilization requirements

It is recommended to use the following disinfectants for 30 minutes:

Case: A customs laboratory adopted a 0.8% sodium hydroxide + 3% o-phenylphenol composite disinfection solution, which can reduce the viral load on the environmental surface by 5 logarithmic levels


Suggestions for the implementation of the standard

1. Focus of staff training

The following skills training needs to be strengthened:

  1. Sample spill emergency disposal process (according to Clause 7.1)
  2. Autoclave operation verification (maintaining 121℃ for 30 minutes)
  3. Level 3 packaging and transportation specifications (Announcement No. 503 of the Ministry of Agriculture)

2. Key points of quality control

The following quality control records should be established:

  • Daily biosafety cabinet wind speed monitoring records
  • Autoclave effect biological indicator test records
  • Positive sample transfer cold chain temperature records
