Introduction

Technical Background and Evolution of the Standard

This standard, an important supplement to the traditional sedimentation incubation method for diagnosing Schistosomiasis japonicum, innovatively utilizes nylon silk bags (260 mesh) pre-filtration technology. Compared to data from the 2000 edition of the "Schistosomiasis Control Manual," filtration efficiency is improved by 40%, achieving an egg recovery rate of 92% ± 3%, significantly superior to the traditional copper sieve method (75% ± 5%).

Key Equipment Technical Specifications

The light incubator must be continuously adjustable from 1-5000Lx, and the microscope must have a magnification of 40×-400×. Nylon silk bags should conform to the conical structure specified in Appendix B (80mm upper opening/15mm lower opening).


Solutions to Common Problems

Case 1: A laboratory's detection rate was low. Upon investigation, it was found that the tap water exceeded the standard for residual chlorine (0.5mg/L). Dechlorination with sodium thiosulfate increased the detection rate by 27%.

Case 2: A mountainous medical institution encountered protozoan interference during incubation using well water. The issue was resolved by adding 7mg/L of bleaching powder as specified in Appendix A.


Implementation Recommendations

  1. Establish a positive control sample library and set up quality control for each batch of testing
  2. In areas with large temperature fluctuations, it is recommended to equip a constant temperature incubator
  3. Use a black background plate (reflectivity <5%) to improve the clarity of miracidia observation
