Key technological breakthroughs

The new standard adopts a gas chromatograph in combination with a hydrogen flame detector (FID), and optimizes the chromatographic conditions: FFAP capillary column (30m×0.32mm×0.25μm), programmed temperature (100℃ for 10min→10℃/min→200℃ for 2min), which significantly improves the separation of propenyl, butenyl and pentenyl isothiocyanates.


Analysis of implementation points

Key points for the preparation of mustard enzyme

According to GB/T5520, mustard seeds with a germination rate of >85% were screened, defatted with petroleum ether 10 times to reduce the fat content to <2%, crushed and passed through a 0.28mm sieve, and stored at -18°C with a shelf life of 6 months. Notes during preparation:

  • Ambient temperature ≤30℃
  • Crush in batches to prevent overheating
  • Activity verification needs to be performed for each batch

Test process optimization

  1. Sample preparation: 2.0g sample + 0.5g mustard enzyme + 5mL pH7.0 buffer + 10mL internal standard solution
  2. 200 times/min oscillation extraction for 2h
  3. 5000r/min centrifugation for 10min and then filtration
  4. Gas chromatography analysis (injection volume 1μL, split ratio 5:1)

Standard implementation recommendations

Laboratories should focus on:

  • The mixed standard solution of isothiocyanates needs to be stored at -18℃ and has a shelf life of 3 months
  • Regularly verify the separation efficiency of the chromatographic column (relative retention time deviation ≤±2.5%)
  • Precision control: RSD≤15% when ≤100mg/kg, RSD≤10% when >100mg/kg
