Introduction
Interpretation of Part 15 of GB/T15670.15-2017 Toxicological Test Methods for Pesticide Registration
This standard specifies the basic principles, methods and requirements for the in vivo mammalian bone marrow polychromatic erythrocyte micronucleus test, which aims to detect whether the test substance causes genotoxicity.
| Standard Dimensions | GB/T15670-1995 | GB/T15670.15-2017 |
|---|---|---|
| Test Scope | Limited to mouse bone marrow polychromatic erythrocyte micronucleus test | Applicable to pesticide registration, covering a variety of mammals (mice and rats) |
| Experimental Methods | Fixed dose design, no clear limit test requirements | Introduction of dose design principles and limit test specifications |
| Result Analysis | Lack of detailed statistical method description | Clarify methods such as Poisson distribution U test and χ2 test |
Explanation of professional terms
Micronucleus: A fragment of genetic material formed by chromosome breakage during cell division, usually with the same color as the main nucleus.
Polychromatic erythrocytes (PC-E): Immature erythrocytes that can be distinguished from normal chromatin erythrocytes (NC-E) by selective staining.
Implementation suggestions
To ensure the accuracy of the test results:
- Mice are preferred as experimental animals, and rats can be used when necessary.
- Strictly control the exposure dose to avoid exceeding the recommended range of LD50.
- Use Giemsa staining solution and phosphate buffer for slide preparation and staining to ensure consistent observation conditions.
- At least 1000 polychromatic erythrocytes were counted for each animal, and the micronucleus rate was calculated.
