1. Key points of virus isolation technology

Note when using SPF chicken embryos for virus isolation:

  • Sample pretreatment: Down samples need to be homogenized after being immersed in physiological saline
  • Inoculation parameters: 9-11 day old chicken embryos, 0.2mL/embryo allantoic cavity inoculation
  • Blind transmission requirements: Samples without HA activity need to be blindly transmitted for 1-3 generations

Biosafety implementation requirements

According to GB19489 standard, special attention should be paid to:

  1. Sample transportation: Samples must be stored at 2-8℃ if delivered within 24 hours, and frozen at -80℃ for more than 4 days
  2. Personal protection: BSL-2 facilities are required for handling infectious materials
  3. Waste disposal: Experimental waste must be sterilized at 121℃ for 30 minutes
