Introduction
Core changes in the standard revision
| Dimensions | 2020 Edition | 2025 Edition |
|---|---|---|
| Scope of application | Highly pathogenic avian influenza only | Expanded to all types of avian influenza |
| Test objects | Poultry tissues | New samples such as down/feather/cowhide |
| Nature of the appendix | IVPI determination is informative | Adjusted to a normative appendix |
| Technical Integration | 6 independent standards | Unified integration into this standard |
Analysis of key diagnostic technologies
1. Key points of virus isolation technology
Note when using SPF chicken embryos for virus isolation:
- Sample pretreatment: Down samples need to be homogenized after being immersed in physiological saline
- Inoculation parameters: 9-11 day old chicken embryos, 0.2mL/embryo allantoic cavity inoculation
- Blind transmission requirements: Samples without HA activity need to be blindly transmitted for 1-3 generations
2. RT-PCR detection specifications
| Gene target | Primer pair | Product size | Judgment standard |
|---|---|---|---|
| M gene | MU/ML | 229bp | Basic positive judgment |
| H5 subtype | H5-U/H5-L | 392bp | Need sequencing verification |
| H7 subtype | H7-U/H7-L | 501bp | Need sequencing verification |
Biosafety implementation requirements
According to GB19489 standard, special attention should be paid to:
- Sample transportation: Samples must be stored at 2-8℃ if delivered within 24 hours, and frozen at -80℃ for more than 4 days
- Personal protection: BSL-2 facilities are required for handling infectious materials
- Waste disposal: Experimental waste must be sterilized at 121℃ for 30 minutes
Clinical judgment criteria
Characteristics of suspected cases: Laboratory confirmation is required if any of the following items are met
- Respiratory symptoms with neurological abnormalities
- Sudden drop in egg production and the appearance of deformed eggs
- Necrotic foci of the pancreas + glandular stomach bleeding were found in autopsy
Technical comparison of the new and old standards
Main enhancements of the 2025 version:
- Breadth of detection: New primers for detecting subtypes such as H3/H9
- Method sensitivity: Real-time fluorescence RT-PCR Ct value determination is more accurate
- Species coverage: Improve the detection process for mammals such as dairy cows
Implementation suggestions
- Laboratories should establish a regular calibration system for fluorescence quantitative PCR instruments
- The sample pre-treatment process needs to be optimized for down product testing
- It is recommended to set up two positive and negative controls for each batch of experiments
