Introduction

Interpretation of the core content of the standard

This standard specifies the technical specifications for the testing of mustard (Brassica juncea) variety specificity (Distinctness), uniformity (Uniformity) and stability (DUS), and is applicable to 16 varieties including leaf mustard, stem mustard, root mustard and stalk mustard and their hybrid varieties.

Key Technical Requirements

Propagation Material Requirements

Material Type Seed Amount Clarity≥ Germination Rate≥ Moisture Content≤
Parental Line 100g 98% 85% 8%
Hybrid 100g 98% 90% 8%
Conventional type 100g 98% 90% 8%

Note:The detection method must be carried out in accordance with GB/T 3543.3, GB/T 3543.4 and GB/T 3543.6.

Test Method Specifications

Field Experiment Design

  • Period:At least 2 independent growth cycles
  • Plot Setting:The variety to be tested is planted adjacent to similar varieties, with ≥100 plants per plot, 2 replications
  • Previous Crop Restriction:Cruciferous crop rotation is prohibited

Trait Observation System

Observation Type Code Definition Sample Size
Population Measurement MG Measure a batch of plant organs to obtain population records Entire plot
Individual measurement MS Measure plant organs one by one to obtain individual records 20 plants
Group visual inspection VG Visually inspect a batch of plant organs to obtain group records Entire plot
Individual visual inspection VS Visually inspect plant organs one by one to obtain individual records 20 plants

DUS judgment criteria

Specificity judgment

The variety to be tested must have significant and reproducible differences from all known varieties in at least one trait.

Consistency threshold

Variety type Population standard Acceptance probability 200 samples allow for heteromorphic strains
Conventional varieties/hybrids 4% 95% ≤13 strains
Maintainer line/paternal line 2% 95% ≤7 strains
Sterile line 1% 95% ≤5 strains

Stability presumption

The stability of trait expression is verified by planting the next generation of seeds, and hybrids can be indirectly determined by the stability of their parents.

Key grouping traits

  1. Cotyledon anthocyanin II coloration (trait 1)
  2. Plant branching (trait 3)
  3. Midrib tubercles (trait 24)
  4. Petiole length/leaf length ratio (trait 27)
  5. Leaf clasping characteristics (trait 31)
  6. Stem enlargement characteristics (trait 37)
  7. Enlarged stem tubercles (trait 42, only for stem enlargement varieties)
  8. Axillary bud enlargement (trait 47, only for stem enlargement varieties)
  9. Root enlargement (trait 53)
  10. Bud appearance period (trait 60)

Implementation suggestions

Testing precautions

  • Trait observations must be carried out strictly in accordance with the growth stages specified in Appendix A (Table B.1)
  • Quality traits (QL) and quantitative traits (QN) require different statistical analysis methods
  • For hybrids, the DUS characteristics of parental materials should be tested simultaneously

Fill in technical questionnaire

The applicant must provide the following in detail as required by Appendix C

  • Variety breeding process and pedigree information
  • Trait difference comparison table with similar varieties
  • Description of suitable planting areas for the variety
  • Representative plant photos (observed traits must be marked)
