Introduction

1. Scope

This standard applies to the quarantine and identification of the pathogen of rapeseed stem canker in rapeseed and its host seeds and plants.

2. Instruments and Equipment

3. Reagents and Culture Medium

The main reagents include 1.0% sodium hypochlorite, agar powder, potatoes, glucose, etc. The culture medium is divided into potato dextrose agar (PDA) and acid potato dextrose agar (APDA).

4. Quarantine identification method

Seed detection process

  1. Pick abnormal seeds for isolation and culture.
  2. After sieving, take samples for initial PCR test.
  3. Positive samples are further isolated and cultured to confirm the results.

5. Identification characteristics

Identification is combined with morphological and molecular biological methods (such as PCR detection) to ensure accurate identification of pathogens.

6. Implementation suggestions

  • The laboratory needs to be equipped with instruments and reagents specified in the standards.
  • Operators should receive professional training.
  • Calibrate equipment regularly and update test methods.
