Introduction

National Standard of the People's Republic of China GB/T 34727—2017 Test of Turtle Germplasm

Background and Significance of Standard Formulation

GB/T 34727—2017 Test of Turtle Germplasm is an important aquatic industry standard, mainly for the detection and identification of turtle germplasm. The standard was issued in 2017, proposed by the Ministry of Agriculture of the People's Republic of China, and managed by the Freshwater Aquaculture Sub-Technical Committee of the National Technical Committee for Aquatic Standardization.

With the rapid development of my country's aquaculture industry, turtles, as important economic species, play an important role in scientific research, breeding and international trade. In order to standardize the test methods of turtle germplasm and ensure the scientificity and accuracy of germplasm identification, this standard came into being.

Test items Biological characteristics Measurement methods Genetic characteristics
Morphological characteristics Body shape, limb shape, skin structure, head characteristics, number of scutes on the carapace and plastron, etc. Fish plate measurement method, refer to GB/T 18654 series standards. Genomic identification through karyotype analysis and isozyme electrophoresis.
Age identification Observation of annual rings of carapace vertebral shields or costal shields. Record the sparse and dense ring patterns under the dissecting microscope to determine the age of the turtle. Study on the association between chromosome karyotype analysis and germplasm characteristics.
Growth and reproduction Carapace length, width and weight data of different age groups. Classification and statistics of carapace length, width and weight of different age groups of turtles in Table 3. Analysis of the influence of genetic characteristics on growth and reproduction.

Professional terms and practical applications

In the determination of turtle germplasm, the number of scutes on the carapace and plastron is a key indicator. For example:

  • Neck scutes: a small scutellum in the middle of the front edge of the carapace.
  • Vertebral scutes: a vertical row of scutes in the middle of the carapace, usually 5 scutes.

In actual applications, precise measurements are performed using instruments such as dissecting microscopes and vernier calipers. These measurement data are combined with genetic characteristics (such as chromosome karyotype and isozyme electrophoresis results) to form a comprehensive germplasm identification system.

Comparative analysis of standard frameworks

Dimensions GB/T 34727—2017 International similar standards
Detection scope Covering biological characteristics, internal structure, genetic characteristics, etc. Focus on ecological adaptability and genetic diversity.
Technical methods Use sampling method and standardized detection process. In-depth analysis is carried out by combining molecular biology techniques.
Data requirements Measure accurately to the millimeter level to ensure data comparability. Emphasis is placed on high-throughput data analysis.

Implementation recommendations

  1. In the process of turtle breeding, regular germplasm determination is carried out according to standards.
  2. Use standardized measurement tools (such as fish measuring board and vernier caliper) to ensure data accuracy.
  3. Combined with genetic testing techniques (such as isozyme electrophoresis and chromosome karyotype analysis) to improve the scientific nature of germplasm identification.
  4. Establish a turtle germplasm database to provide a reference for subsequent research and management.
