Introduction

1. Background and significance of the standard

GB/T 34748—2017 is China's first national standard for genomic DNA microsatellite analysis of aquatic germplasm resources, which aims to standardize the experimental methods and data analysis processes in this field. The standard was proposed by the Ministry of Agriculture and managed by the National Technical Committee for Aquatic Products Standardization. The main drafting unit is the Heilongjiang Fisheries Research Institute of the Chinese Academy of Fishery Sciences.

3. Implementation suggestions and operation guidelines

3.1 Sample collection and storage

According to GB/T 18654.15, the sample size should be more than 40. The sampling location, time and method should be recorded in detail, and the "Microsatellite Analysis Record Form of Genomic DNA of Aquatic Organisms" (Appendix C) should be filled in. After the sample is collected, it should be stored immediately in accordance with the specifications to avoid degradation.

3.2 Reagent and instrument preparation

  • Use Genomic DNA Extraction Kit or verify a reliable extraction method.
  • Prepare microsatellite marker primers (100μmol/L) and store them properly.
  • Prepare PCR amplification instruments (such as vertical electrophoresis tank and high voltage constant power electrophoresis instrument).
