Introduction

Interpretation of the National Standard of the People's Republic of China GB/T 35539—2017

Analysis of the Standard Formulation Background and Technical Evolution of Restriction Endonuclease BamHI

BamHI is an important restriction endonuclease and its application in genetic engineering and molecular biology research is irreplaceable. With the rapid development of gene editing technology, the demand for BamHI is increasing. The formulation of national standards aims to regulate its production and quality control to ensure its efficient application in scientific research and industry.

Comparison of Standard Frameworks

Dimensions Standard Requirements Technical Parameters
Enzyme Activity (U/mL) {≥10000 U/mL} BamHI activity unit definition: The amount of enzyme required to completely decompose 1 μg DNA within 1 h in a 50 μL reaction system at 37°C is 1 activity unit.
Impurity Control Should not contain other endonucleases and exonucleases Enzyme purity is ensured by multiplex PCR detection technology (Appendix A) and electrophoresis analysis.
Storage conditions Temperature below -20°C Avoid loss of enzyme activity due to high temperature and ensure a shelf life of 3 years.

BamHI detection method interpretation and application case

Appendix A specifies the quantitative detection method of BamHI enzyme activity in detail. Through PCR and electrophoresis technology, it is possible to accurately determine whether the enzyme activity meets the standard requirements. For example, in a multiplex PCR experiment, if 1 μg of DNA is not completely cut, it indicates that the enzyme amount is insufficient, otherwise it indicates that the enzyme amount meets the standard.

Implementation suggestions

  • Selection principles:Choose BamHI products that meet the standard requirements according to experimental needs, and give priority to products with high enzyme activity and low impurity interference.
  • Quality Control:Establish a strict testing process, including enzyme activity verification and impurity detection, to ensure stable product quality.
  • Safe Storage:Store at -20°C as required by the standard, and regularly monitor changes in enzyme activity.
