Introduction
National Standard of the People's Republic of China GB/T 36810—2018 Quarantine Identification Method for Strawberry Fusarium wilt
Background and Significance of the Standard
Strawberry Fusarium oxysporum f. sp. fragariae is a quarantine disease that seriously harms strawberry cultivation. This standard was proposed and coordinated by the National Technical Committee for Plant Quarantine Standardization, and the main drafting unit was the Tianjin Entry-Exit Inspection and Quarantine Bureau of the People's Republic of China.
Detection principles and method framework
| Dimensions | Basis for quarantine identification of pathogens | Requirements for molecular biological identification |
|---|---|---|
| Symptom inspection | Observe the symptoms of poor plant growth, dwarfing, weakness and rot, the lower leaves turn purple and wilt, and the heart leaves turn yellow-green or yellow. | - Take suspicious materials for surface disinfection and then isolate and culture - Observe the colony morphology and spore characteristics |
| Pathogen isolation and culture | Use PSA or PDA culture medium and observe the colony characteristics after culturing at 25°C for 48 hours. | - PCR amplification was performed using specific primers FofraF/FofraR - The target fragment was 239bp |
| Molecular biological identification | Extract strain DNA, perform PCR reaction and perform electrophoresis detection. | - PCR reaction system includes EmeraldAmp Max PCR Master Mix - Amplification products are analyzed by electrophoresis in 2% agarose gel |
Instrument and reagent requirements
The following are the main instruments and reagents required for the detection process:
microscope, clean bench, biological incubator and other instruments and equipment, as well as EDTA, SDS, Tris-HCI, primers FofraF/FofraR and other reagents.
Analysis of pathogen identification characteristics
| Identification index | Characteristics of strawberry wilt pathogen | Differences between similar species |
|---|---|---|
| Culture characteristics | The colonies on the PSA plate are raised, white, dense and flocculent, and the back is purple. | - Fusarium equiseti: The hyphae are velvety, and the surface of the substrate is light brown - Fusarium sambuciumu: Produces a large number of orange-red conidia |
| Morphological characteristics | Small conidia are oblong, and large conidia are sickle-shaped. | - Fusarium equiseti: spindle-shaped or sickle-shaped, mostly with 3 septa - Fusarium sambuciumu: papillary apical cells |
Implementation suggestions and precautions
1. Strengthen the quarantine of strawberry nurseries to ensure that there is no entry of pathogens.
2. Standardize the soil treatment process and use high-pressure steam sterilization and other methods to eliminate pathogens.
3. Improve farmers' awareness of strawberry wilt and promote healthy cultivation techniques.
