Analysis of key technical points
Key indicators for morphological identification
The standard clearly states that the typical characteristics of Blacktail Anopheles nigrophila are: the lower lobe of the deeply forked caudal fin is long, the third anal spine is well developed, the swim bladder has 2-3 chambers and the length of the rear chamber is 1.8-2.9 times that of the front chamber. These characteristics can be distinguished from closely related species.
Genetic testing specifications
Chromosome testing requires the preparation of 7% polyacrylamide gel and vertical plate electrophoresis using a Tris-glycine buffer system (pH8.3). Isozyme detection requires the collection of lens tissue from more than 10 samples, and the supernatant is sampled after centrifugation.
Implementation recommendations
- Sampling should cover different age groups, and stratified sampling should be performed according to GB/T 18654.2
- The scale method is recommended for age identification, and attention should be paid to the identification of reproductive scales and regenerated scales
- Isozyme detection requires strict control of electrophoresis conditions (280V voltage regulation for 10min + 228V constant voltage for 4h)
Background of standard evolution
This standard was formulated by the Wuhan Academy of Agricultural Sciences and fills the gap in the germplasm standards of the subfamily of bream in my country. Compared with the GB/T 18654 series of standards, Blacktail Anchovy-specific trait indicators and genetic marker detection procedures have been added.
