Morphological characteristics
Collichthys lucidus has an oblong body shape, with the following characteristics: prominent occipital ridges, gray-black front end of gill cavity, and pale yellow fins. Among the measurable traits, the ratio of body length to tail peduncle length is 3.806±0.311, and the head length to eye distance is 4.687±0.522.
Molecular genetics
The standard stipulates that the mitochondrial COI gene detection needs to use primers COLF/COLR, PCR reaction system 50μL, annealing temperature 50℃. Karyotype analysis shows 2n=48t, NF=48, and the chromosome karyotype is shown in standard Figure 2.
Implementation and application suggestions
Testing process optimization
1. Sample collection: According to the requirements of GB/T 18654.2, samples during the breeding period need to be differentiated by sea area
2. Age identification: Otolith annual ring analysis is preferred
3. Molecular testing: It is recommended to add bidirectional sequencing to verify the COI sequence
Standard evolution analysis
This standard integrates morphological and molecular genetic indicators for the first time. Compared with the earlier version:
- Added the anatomical characteristics of 21-22 pairs of swim bladder limbs
- Standardized chromosome karyotype formula expression (2n=48t)
- Appendix A specifies the conditions for DNA extraction and PCR amplification in detail
