Key operation points
The sample pretreatment used liquid nitrogen cryo-grinding technology, combined with 0.1% formic acid aqueous solution extraction, which significantly improved the recovery rate of hydrophobic tryptamines (83.5-109.7%). The addition of 20mmol/L ammonium acetate buffer system in the mobile phase effectively improved the peak shape of 5-methoxytryptamine derivatives.
Mass spectrometry parameter optimization strategy
In view of the indole parent nucleus characteristics of tryptamines, the standard recommends 5500V ion source voltage and specific collision energy (20-67eV). For example, 5-MeO-DiPT needs to set DP45V/CE20eV, while 4-Acetoxy-DiPT needs DP45V/CE46eV, which reflects the parameter optimization of structural differentiation.
Implementation suggestions
- Regularly verify the stability of psilocybin-d4 and psilocybin-d10 internal standards
- Pay special attention to the column balance of 4.5-6.5min during the gradient elution phase
- It is recommended to add 0.05ng/mg quality control points for each batch of samples
