Introduction
Technical background of the standard
This standard is formulated to meet the quarantine and identification requirements of Amblyomma compressum Macalister, 1872, an important quarantine pest belonging to the family Ixodidae of the order Ixodida. This species has been historically known by three synonyms, including Haemalastor compressum.
Core identification features
| Location | Female characteristics | Male characteristics |
|---|---|---|
| Pseudo-head base | 6.8mm wide, obvious lateral angles | 5.0-5.2mm wide, blunt lateral angles |
| Pedals | Second segment has a long dorsal spur | Second segment has a dentate process on the ventral side |
| Shield | Oval, with little enamel | Fully covered, with special enamel pattern |
Typical identification case
In 2018, a suspected individual was detected from the surface of imported monitor lizards at Shenzhen Port. Based on the female anal groove characteristics described in Article 7.3.1 of this standard (circumferential and U-shaped at the front) and the key indicator of valve plate comma shape, it was finally confirmed as the first interception at a Chinese port.
Key points of technical implementation
- Sample processing: Use 75% ethanol to fix, and keep the specimen horizontal when measuring under a stereomicroscope
- Identification of closely related species: Focus on comparing the scutellum pattern and basal gland structure of the Javan Ornithodoros tick (A. javanense)
- Molecular verification: It is recommended to sequence and align the COI gene, and the sequence similarity should be ≥98%
Analysis of standard evolution
Compared with the 2009 version of the GB/T 1.1 basic standard, this standard establishes a complete morphological indicator system for Compressed Ornithodoros tick for the first time:
- Added 12 microscopic measurement data (such as the length-to-width ratio of the second segment of the female pedipalp is 1.7:1)
- The threshold for determining the subcircular structure of the valve plate is clarified
- The requirements for the extension angle of the foot IV during specimen preparation are standardized
