Compared with the 48-72-hour detection cycle of the traditional culture method, mass spectrometry technology improves efficiency by more than 90% while maintaining >95% identification accuracy.

Key parameters of mass spectrometry technology

Experimental data show that when α-cyano-4-hydroxycinnamic acid (CHCA) is used as the matrix, the best signal-to-noise ratio can be obtained at an accelerating voltage of 20kV. Typical characteristic peaks include:

  • Ribosomal protein S10 (m/z 5250)
  • DNA binding protein (m/z 10300)

Implementation suggestions and precautions

1. Quality control: BTS standards must be used for calibration before each test, and the mass-to-charge ratio error must be <0.03%
2. Biosafety: All cultures must be sterilized at 121℃ for 30 minutes
3. Result verification: Matching scores ≥2.3 can be reported directly, and 1.7-2.2 must be verified in combination with GB/T 14926.13
