This standard specifies a dual method system of morphological identification and DNA barcode identification for Dendrobium nobile Lindl., which is applicable to the import and export inspection and quarantine of Chinese medicinal materials.

Molecular biology identification process

  1. DNA extraction: CTAB method or commercial kit
  2. PCR amplification: ITS gene primers (848bp)

Technological evolution and standard value

This standard uses the ITS sequence as the legal identification basis for the first time, which solves the limitations of traditional morphological identification in dried medicinal materials. The 21 similar species keys provided in the appendix can effectively distinguish easily confused species such as Dendrobium tassellensis and Dendrobium curcumae.

  • It is recommended to compare and identify with the morphological characteristics in Appendix B
  • The DNA sample should meet the quality requirement of OD260/280 ratio of 1.7-2.0
  • The standard strain with GenBank accession number MT940440.1 is recommended for positive control
