The standard uses a microwave digestion instrument for sample pretreatment, using a nitric acid-hydrogen peroxide system at a high temperature of 210°C to completely decompose the organic matrix. The arsenic element needs to be pre-reduced to As(III) by a thiourea-ascorbic acid system, while the mercury element exists directly in the form of Hg²⁺.
Optimization of key instrument parameters
Atomic fluorescence spectrometer parameter settings reflect differentiation:
- Arsenic detection uses 300V negative high voltage and 60mA lamp current
- Mercury detection needs to be reduced to 280V negative high voltage and 30mA lamp current

Implementation suggestions
Note in actual operation:
- The microwave digestion procedure strictly follows the step-by-step heating mode of 15min to 170℃ → 8min to 210℃ → maintain for 20min
- Before arsenic determination, it is necessary to ensure that the valence state conversion is completed by standing at 20±10℃ for 20 minutes
- The mercury standard solution should be prepared and used immediately to prevent adsorption loss
