Introduction
GB/T 31800-2015 Standard Overview
This standard specifies the immunological and molecular biological quarantine identification methods for plum pox virus, and is applicable to the quarantine identification of plum pox virus in plant materials including asexual propagation materials, tissue culture seedlings, seeds, flowers and fruits.

Implementation recommendations for quarantine and identification technology of plum pox virus
Step 1: Preliminary inspection of symptoms
- Observe the symptoms of plant leaves, flowers and fruits, such as discoloration, ring spots, deformities, etc.
- Record suspicious samples and conduct subsequent laboratory tests.
Step 2: Double Antibody Enzyme-Linked Immunosorbent Assay (DAS-ELISA)
- Suitable for rapid initial screening, negative results can be directly judged as undetected.
- Positive results need to be further verified by RT-PCR.
Step 3: Molecular Biology Detection
- Conventional RT-PCR and real-time fluorescence RT-PCR are suitable for the confirmation stage and have high specificity and sensitivity.
- Real-time fluorescence RT-PCR can also perform quantitative analysis of viral load.
Step 4: Strain Identification
- Based on the test results, virus strain identification can be performed if necessary to distinguish different geographical populations and genotypes.
- Refer to the primer sequences and amplification conditions in Appendix E to ensure accuracy and repeatability.
