《GB/T 34408-2017天然皮革牛、羊、猪源性成分定性PCR检测方法》由TC374(全国质量监管重点产品检验方法标准化技术委员会)归口,主管部门为国家标准化管理委员会。
Introduction
1. Background and significance of the standard
GB/T 34408—2017 is my country's first qualitative test method standard for cattle, sheep and pig-derived components in natural leather, filling the gap in related fields at home and abroad. The standard was proposed by the National Technical Committee for Standardization of Key Product Inspection Methods for Quality Supervision and was issued and implemented in 2017.
With the rapid development of the leather industry, mixed sources and counterfeiting and shoddy products have become increasingly prominent. This standard provides important technical support for ensuring product quality and regulating market order.
2. Comparison of standard frameworks
| Standard dimensions | GB/T 34408—2017 | International reference standards | Other domestic standards |
|---|---|---|---|
| Scope of application | Qualitative detection of buffalo, cattle, goat, sheep and pig-derived components in natural leather | Detection methods covering multiple animal-derived components | Mainly for single species detection |
| Technical route | Target gene sequence analysis based on PCR amplification | Combination of multiple molecular biology techniques | Reliance on traditional detection methods |
| Detection efficiency | High efficiency and sensitivity, able to detect multiple components simultaneously | Time-consuming, requiring multiple experiments | Long detection cycle and high cost |
3. Interpretation of key technical points of the standard
3.1 Reagents and materials
The standard specifies in detail the preparation methods and quality requirements of various reagents, including:
- Analytical balance: accuracy ±0.01g
- PCR thermal cycler: used to amplify target genes
- Electrophoresis instrument: detect PCR products
All reagents must meet the corresponding quality standards to ensure the accuracy of the experiment.
3.2 Testing steps
The core process includes:
- Decolorization: Dark leather needs to be decolorized first to avoid interference with subsequent testing.
- DNA extraction: Purified DNA samples are obtained through multiple steps of centrifugation and washing.
- PCR amplification: Specific primers are designed for the target species and amplified.
- Electrophoresis: Amplification results are observed through agarose gel electrophoresis to determine whether they are positive.
4. Implementation suggestions
To ensure the effective implementation of the standard:
- Strengthen the training of testing personnel and familiarize them with PCR technology and experimental operations.
- Establish a complete quality control system and calibrate instruments and equipment regularly.
- Promote the construction of standardized testing laboratories and unify the configuration of reagents and equipment.
