Introduction

Introduction

Hostavirus X (HVX) is a disease that seriously affects ornamental plants in gardens. In order to standardize its quarantine and identification methods, the National Standard of the People's Republic of China GB/T 35330-2017 came into being. This standard combines a variety of detection technologies to ensure accurate determination of whether a sample carries the HVX virus in different scenarios.

Comparison of Standard Frameworks

Dimensions GB/T 35330-2017 SN/T 2964 SN/T 3457
Scope of Application Quarantine and Identification of Hosta Virus X General Methods for Plant Virus Detection Molecular Biological Detection of Plant Viruses
Core Technologies DAS-ELISA, RT-PCR, Real-time Fluorescence RT-PCR, RT-LAMP Virus Isolation and Identification Genome Detection Technology
Update frequency Revised regularly and adjusted according to the development of the disease Revised every 5 years Revised every 3 years

Key technology analysis

DAS-ELISA test:By specifically binding the HVX virus with antibodies, rapid initial screening is achieved. This method is easy to operate and suitable for large-scale sample testing.

RT-PCR technology:Based on the genomic characteristics of HVX, specific primers are designed for amplification. This method has high sensitivity, but the reaction conditions must be strictly controlled.

Real-time fluorescence RT-PCR:Combined with fluorescently labeled probes, quantitative detection is achieved. This technology is efficient and accurate, and suitable for high-throughput sample analysis.

RT-LAMP detection:Uses loop-mediated isothermal amplification technology to complete the detection at a constant temperature. This method is particularly suitable for on-site rapid detection with limited resources.

Instruments and reagents

The following are the main instruments and reagents involved in the standard:

  • Electronic balance: Sensitivity 0.001g
  • PCR instrument: used for amplification reaction
  • Enzyme reader: detect ELISA reaction results
  • High-speed refrigerated centrifuge: separate virus particles

Implementation recommendations

To ensure the effective implementation of the standard, the following recommendations are made:

  1. Train quarantine personnel and familiarize them with the operating procedures of various detection technologies.
  2. Establish standardized laboratories and equip them with necessary instruments and equipment.
  3. Regularly update test kits to ensure detection sensitivity and specificity.
  4. Strengthen international cooperation and share HVX virus monitoring data.
