Analysis of the core content of the standard

This standard systematically standardizes the entire process of sample pretreatment for microbial research in wastewater treatment systems, with a focus on three major technical modules:

  • On-site treatment specifications: clarify the collection and fixation methods of different samples such as water samples, activated sludge, and biofilm
  • Bacterial strain isolation technology: establish a graded separation and culture system for bacteria/actinomycetes/fungi
  • Metagenomic extraction: specify the quality control indicators for CTAB method DNA extraction

Typical application scenarios

A sewage treatment plant uses Membrane bioreactor process, special attention should be paid to:

  1. The biofilm sample needs to be completely scraped from the surface of the polyvinylidene fluoride membrane with a sterile blade
  2. The OD260/280 ratio needs to be controlled between 1.8-2.0
  3. Fixed samples should use 50% ethanol instead of formaldehyde to avoid DNA damage
