《GB/T 43271-2023木材鉴定 DNA条形码方法》由TC41(全国木材标准化技术委员会)归口,TC41SC1(全国木材标准化技术委员会基础标准分会)执行,主管部门为国家林草局。

Introduction

1. Background and significance of standard formulation

The current wood anatomical method can usually only identify to the 'genus' level, which is difficult to meet the needs of international trade law enforcement and domestic market supervision and management for wood 'species' level identification. The DNA barcode method provides a reliable technical basis for the accurate identification of wood.

2. Comparison of standard frameworks

Standard dimensions GB/T 43271—2023 International Technical Specification for Timber Identification
Scope of application Applicable to the identification of species of wood and its products Mainly for high-end wood materials
Core technology DNA barcode analysis, PCR amplification technology Traditional anatomy combined with chemical analysis
Detection accuracy Species level identification Mainly genus level

3. Key technologies and instruments

PCR instrument: used to amplify specific DNA fragments, key parameters include reaction temperature, number of cycles, etc.

Electrophoresis instrument: detect the size of PCR products through agarose gel electrophoresis and evaluate the amplification effect.

Sequencer: used to obtain DNA sequence information, it is the core equipment for identification.

4. Experimental steps and judgment methods

The experimental steps include sample collection, DNA extraction, PCR amplification, electrophoresis detection and sequence analysis. The judgment method is based on sequence consistency and molecular phylogenetic tree construction to ensure the accuracy of the identification results.

5. Requirements for identification report

  • Include photos and descriptions of wood samples
  • Detailed record of test methods and judgment basis
  • Clear identification results and attach relevant explanations

6. Implementation suggestions

To ensure the effective implementation of the standard, it is recommended to:

  1. Strengthen laboratory capacity building and equip with advanced instruments and equipment.
  2. Establish an authoritative DNA barcode reference database.
  3. Formulate unified quality control standards.
  4. Promote international technical exchanges and cooperation.
