This standard is a quarantine identification method for Opjiostoma zaagereri, which originated from the devastating damage of the pathogen to pine and fir plants in North America. In 2012, the Chinese quality inspection system established a special inspection standard for the first time, filling the technical gap in the quarantine of conifer root diseases in China.

Field sampling specifications

It is necessary to collect 20-30cm wood segments together with the vector insects (such as black fat beetles), and focus on observing three types of section characteristics:

  1. Cross-section: Look for black crescent-shaped lesions
  2. Radial section: Check for longitudinal black stripe lesions
  3. Chordal section: Confirm conidiophores in the insect tunnels

Laboratory identification process

Use the double verification method:

  • Microscopic examination: Observe the characteristics of hyphae penetrating the marginal pits
  • Cultivation: PDA selective medium (containing 0.2g/L actinomycin) at 18℃ for 3 days

Standard application case

In 2018, Tianjin Port detected suspected cases in imported Douglas fir logs through:

  1. Comparison of conidiophore morphology (30-940μm×10-12μm)
  2. Verification of the actinomycin resistance of mycelium
  3. PCR gene sequencing confirmation

It was finally determined to be the Ophiostoma wageneri var. pseudotsugae variant, successfully blocking the introduction of the epidemic.
