Standard Overview and Technical Background

SN/T 4715-2016 is my country's first industry standard for the full lifecycle management of weed specimens in the entry-exit inspection and quarantine sector. It is based on GB/T 1.1-2009, the Guidelines for Standardization. This standard, for the first time, combines DNA barcoding technology with traditional specimen preparation requirements, reflecting the development trends of modern plant taxonomy.

2.1 Specimen collection specifications

2.2 Specimen processing technology

Special provisions of the standard DNA materials require a two-step treatment: pre-fixation with 70% ethanol followed by rapid drying on silica gel to ensure DNA fragment integrity. Comparative experiments have shown that this method can increase DNA extraction success rates by over 40%.


Preservation management requirements

4.2 Preservation environment control

  • Herbara specimens: temperature 15-25℃, humidity 40-50%
  • DNA materials: long-term storage at -20℃ or ultra-low temperature storage in liquid nitrogen
  • Specimen cabinets must be equipped with insect-proof camphor preparations

Implementation suggestions

  1. When collecting in the field, be equipped with a portable dryer and a DNA rapid preservation kit
  2. Establish a specimen-gene dual numbering system
  3. Regularly monitor the temperature and humidity fluctuations of the storage environment
