Technical Background of the Standard
SN/T 4727-2016 is my country's first special testing standard for pseudo-root-knot nematode (Nacobbus aberrans), filling the gap in quarantine nematode identification technology.

Molecular detection technology
The standard adopts the PCR primer system of SN/T 2757:
- Forward primer: 5'-CGTGGAATGATCAGAACGC-3'
- Reverse primer: 5'-TCCCAAAGACCTTCATTCTTC-3'
Host Plant Range
Ruboid knot nematodes can infect over 100 plant species across 24 families, primarily including:
- Solanaceae: potato, tomato, pepper
- Cucurbitaceae: cucumber, pumpkin
- Cruciferae: cabbage, rapeseed
- High-risk quarantine hosts: beet, carrot
Recommendations for Implementation of the Standard
1. Sample Storage: Refrigerate at 4°C for no more than 3 days; long-term storage requires freezing at -20°C.
2. Morphological identification requires measurement of at least 20 morphological parameters.
3. Molecular testing should include a positive control (ATCC 49236).
4. Quarantine treatment: soil fumigation is required if an epidemic is found (methyl bromide 40g/m³ is recommended)
