Key steps of colorimetry

  1. Use sulfanilic acid (C6H7NO3S) to react with nitrite for diazotization
  2. Add N-1-naphthylethylenediamine hydrochloride to generate a purple-red compound
  3. Measure the absorbance at a wavelength of 538nm

Special requirements for ion chromatography

  • Requires AS19 chromatographic column (50mm×4mm)
  • Eluent gradient: 6-70mmol/L KOH solution
  • Injection volume: 250μL

Standard implementation recommendations

Sample pretreatment precautions

For high-protein feed (such as fish meal), it is recommended to use 70℃ water bath extraction for 15 minutes and then cool immediately to avoid protein interference. Liquid samples need to be filtered through a 0.22μm filter membrane.

Quality control requirements

Each batch of tests should include: blank control, spike recovery (80-110%) and parallel samples (RSD≤10%).


Technical development background

This standard replaces part of the GB/T 5009.33-2008 method. The main improvements include: reducing the detection limit by 50%, adding a solid phase extraction cleanup step, and introducing gradient elution chromatography technology.
