Introduction
Technical background of the standard
This standard replaces SN/T 3011.1-2011. Aiming at the species identification needs of products derived from sika deer (Cervus Nippon), it introduces real-time fluorescence PCR technology to improve detection sensitivity (up to 100±50 ng DNA) and specificity. Result interpretation criteria
Positive judgment: When the Ct value is ≤35 and the control is normal, the sika deer component is detected. When the Ct value is in the range of 35-40, it needs to be retested for confirmation.
The typical amplification curve should show S-shaped characteristics, and the probe uses the FAM-TAMRA fluorescent labeling system.
Implementation recommendations
- The laboratory should regularly verify the primer specificity (refer to Appendix A sequence)
- It is recommended to set up triple controls (positive/negative/blank) for each batch of testing
- For deep-processed samples, the DNA extraction plan needs to be optimized
