Technical background and evolution of the standard
As the fifth part of the SN/T 5228 series, this standard marks the leapfrog development of my country's detection technology for pathogenic microorganisms in exported food from traditional culture method to matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS). Compared with the traditional method of ISO/TS 21872-2, the detection cycle is shortened from 5-7 days to within 24 hours.

Detailed explanation of key points of operation
Sample pretreatment
The ethanol-formic acid two-step extraction method is used: first fix the cell structure with 90% ethanol, and then dissolve the characteristic proteins with 70% formic acid. In actual application, the Beijing Customs Laboratory found that shellfish samples need to add 10 minutes of ultrasound-assisted extraction.
Instrument calibration
The mass-to-charge ratio calibration must be performed using BTS standard, and the error is required to be <0.03%. It is recommended to recalibrate after every 20 samples to ensure data accuracy.
Result judgment standard
The matching score adopts a three-level judgment system:
• ≥2.300: species level identification is reliable
• 1.700-2.299: needs to be confirmed in combination with biochemical tests
• <1.700: re-cultivation and testing is recommended
Implementation suggestions
- Laboratories need to establish biosafety emergency plans in accordance with GB 19489
- Regularly update the mass spectrometry database (it is recommended to upgrade every quarter)
- This method is preferred for seafood samples
