Interpretation of the core technology of the standard
This standard adopts the 3M MDS Listeria monocytogenes Molecular Detection Kit, which is based on the loop-mediated isothermal nucleic acid amplification (LAMP) technology and achieves specific detection by identifying 6 independent regions of the target sequence unique to the genus Listeria monocytogenes. Key operation process
The test procedure is divided into three core stages:
- Sample pretreatment: Use Demi-Fraser medium for 28-30 hours for primary enrichment, and cheese/fish samples require secondary enrichment with Fraser medium
- DNA extraction: Lysis at 100℃ for 15 minutes, and obtain pink DNA template after cooling
- LAMP amplification: Constant temperature reaction at 60℃, real-time fluorescence detection through the conversion of pyrophosphate to ATP
Quality control requirements
Typical application case: A dairy company used this method to detect positive samples, and the actual contamination level was confirmed to be 0.5 CFU/g by GB 4789.30, which was 72 hours earlier than the traditional culture method.
The standard clearly stipulates that double parallel controls must be set up:
- Positive control: RC control is provided in the kit
- Negative control: Sterile Demi-Fraser broth
Implementation suggestions
1. Laboratory partitioning: Strictly distinguish the sample processing area from the amplification analysis area to prevent aerosol contamination
2. Equipment validation: Regularly calibrate the temperature control system of the 3M molecular detector (60±1℃)
3. Consumables management: The lysis solution (LS tube) needs to be stored in the dark at 2-8℃
4. Result interpretation: The test result is valid only when both positive and negative controls meet expectations
