Introduction

Interpretation of the National Standard of the People's Republic of China GB/T 34733—2017

1. Background and significance of the standard

GB/T 34733—2017 "Diagnostic Procedure for Cryptocaryonosis in Marine Fish" is a diagnostic standard for Cryptocaryonosis, a common and serious disease in marine aquaculture fish. The standard was proposed by the Ministry of Agriculture of the People's Republic of China and was coordinated by the National Technical Committee for Aquatic Products Standardization. The main drafting units include the Fujian Freshwater Fisheries Research Institute and the Fujian Aquatic Animal Disease Prevention and Control Center.

With the rapid development of marine aquaculture, Cryptocaryonosis has become one of the important diseases that affect fish health and aquaculture benefits. The formulation of this standard provides a scientific basis for standardizing diagnostic methods and unifying diagnostic processes, which is of great significance to protecting the healthy development of my country's aquaculture industry.

2. Comparison of standard frameworks

Standard dimensions GB/T 34733—2017 Comparative analysis
Scope of application Diagnosis, epidemiological investigation, quarantine and monitoring of Cryptocaryon irritans in marine fish. Comprehensively covers the entire breeding process, focusing on differentiated management of seedlings and adult fish.
Core content Sample collection and processing, diagnostic methods, pathogen confirmation and result determination. Clear process, emphasizing microscopic examination and worm morphological analysis.
Technical basis Based on GB/T1.1-2009 standardization rules, and citing SC/T7014-2006, SC/T7103-2008 and other normative documents. Ensure the scientificity and operability of the diagnostic method.

3. Interpretation of the core content of the standard

3.1 Reagents and instruments

The reagents and instruments required by the standard include:

  • Normal saline (0.85%)
  • Distilled water
  • Fixative solution (4% formaldehyde or 70% ethanol)
  • Giemsa staining solution
  • Optical microscope, dissecting microscope and magnifying glass (30×)

3.2 Epidemic situation and main symptoms

Epidemic scope:Applicable to almost all marine cultured fish, especially to large yellow croaker, oval grouper and other fry.

Time of onset:Mainly concentrated in late May to mid-July (water temperature is about 22°C~28°C) and mid-September to late November (water temperature is about 25°C~19°C).

3.3 Sample collection and processing

Collect more than 10 fish with clinical symptoms from the culture cages or ponds, and the rest shall be handled in accordance with Chapter 6 of SC/T7014-2006.

3.4 Diagnostic method

Observe the body surface and fins of the diseased fish through a dissecting microscope or a magnifying glass, scrape some mucus from the body surface of the diseased fish, cut some fins and gill filaments of the left and right gills, and make water seals for microscopic examination.

4. Implementation recommendations

Preventive measures: Strengthen seedling quarantine, control water exchange frequency, and maintain a stable environment within the appropriate water temperature range (10°C~30°C).

Diagnostic process: Strictly follow the standard requirements for sample collection, processing and microscopic examination to ensure the accuracy of insect morphological confirmation.

Precautions: When using chemical reagents (such as formaldehyde, ethanol), strictly abide by laboratory safety regulations to avoid human contact and environmental pollution.
