Introduction

1. Background and significance of standard formulation

GB/T 34739-2017 "Detection technology of neutralizing antibodies to animal rabies virus" is a standard formulated in accordance with the relevant requirements of the International Terrestrial Animal Health Code (OIE) and in combination with my country's actual situation. This standard is mainly used to evaluate the immune protection of animals against rabies, and provide a scientific basis for vaccine development, immune effect monitoring and epidemiological research.

2. Overview of FAVN detection method

FAVN (Fluorescent Antibody Virus Neutralization Test) is a virus neutralization test based on fluorescent antibodies, which is used to quantitatively determine rabies virus neutralizing antibodies in animal serum. This method has high sensitivity and high specificity and is widely used in veterinary laboratories.

3. Analysis of the test operation process

Steps Specific requirements Key points
Sample collection Aseptically collect ≥2mL blood and let stand at 4℃ for more than 1 hour. Make sure to record animal information to avoid contamination.
Serum treatment Inactivate in a 56℃ water bath for 30 minutes and centrifuge at 4000r/min for 10 minutes. Use secondary pure water to prepare the solution.
Virus titration BHK-21 cell culture, determination of CVS-11 strain TCID50 value ≥10^5 TCID50/mL. The operation must be carried out in a BSL-2 laboratory.

4. Experimental materials and equipment requirements

Reagent requirements:Including analytical reagents, fluorescent labeled antibodies (FITC), cell culture medium (MEM), etc.

Equipment requirements:Equipped with professional instruments such as CO₂ incubator, inverted fluorescence microscope, 8-channel pipette, etc.
