Introduction

1. Background of standard formulation and technological evolution

GB/T 35526-2017 is based on OECD Chemical Test Method No. 441. Through years of verification and research, the reliability and reproducibility of the test are ensured. This standard clearly applies the Hershberger biological screening test to anti-androgen detection for the first time, and optimizes the experimental design in combination with modern laboratory conditions.

Table 1 shows the maximum allowable CV values of different tissues under anti-androgen and androgen effects to ensure the accuracy of the experimental results.


3. Implementation suggestions

Animal selection and management:Preferably use common laboratory rat strains, ensure that there are at least 6 experimental animals in each group, and strictly control the weight difference within ±20%. Castration surgery should be performed 42 days after birth, and 7 days of adaptation are required after surgery.

Dose setting and poisoning route:It is recommended to set up three dose groups, the maximum dose should not exceed 1000 mg/(kg·d), and set up a positive control group (TP or FT) and a negative control group at the same time. Poisoning can be completed by gavage or subcutaneous injection.

Data analysis and quality control:Perform variance analysis on the target tissue mass to ensure that the CV value is within an acceptable range. For abnormal data, it is necessary to check for record errors or experimental technical problems and retest.
