SN/T 4675.8-2016, a key component of the import and export wine inspection standards, specifically specifies the method for determining 5-hydroxymethylfurfural (5-HMF) using a high-performance liquid chromatograph (HPLC). 5-HMF is a Maillard reaction product, and its content can reflect the wine's heat treatment and storage conditions.

Key Operation Procedures

Sample Pretreatment: Take 10.0 mL of alcohol sample and dilute to 100 mL with 10% methanol solution. Filter through a 0.45 μm filter before injection. The standard curve should cover a concentration range of 0.1-5.0 mg/L.

Chromatographic Condition Optimization: Under the conditions of a column temperature of 30°C, a flow rate of 1.0 mL/min, and an injection volume of 10 μL, the retention time of 5-HMF should be stable at 10.6 ± 0.3 minutes.


Method Validation Data

Standard validation showed that: within the concentration range of 1.0-20.0 mg/L, the spiked recoveries in different matrices were 92.8%-109%, with relative standard deviations (RSDs) less than 5%. The method detection limit was 1.0 mg/L, and the limit of quantification was 2.1 mg/L.

  2. Samples should be stored away from light and tested within 24 hours.
