SN/T 4726-2016 is the quarantine and identification industry standard formulated by my country for Adoxophyes orana (apple tormenter). As an important cross-border agricultural pest, the identification technology of this species has undergone three major iterations:

Analysis of core identification characteristics

Morphological identification of adults

According to clause 7.2.1 of the standard, adults of Adoxophyes orana have the following key characteristics:

  • Forewings are 13-23mm, with a yellowish-brown to tan base
  • Wings have typical reticulated patterns and three silvery horizontal bands
  • Hindwings are off-white with distinct cilia

Actual case: intercepted at Shanghai Port in 2018

By comparing the separation angle of veins R4 and R5 (the standard requirement is ≤45°), it was successfully distinguished from the closely related species Clepsis spectrana


Technical specifications for molecular detection

Appendix E of the standard specifies the molecular identification process:

  1. DNA extraction: chest muscle tissue is processed using the CTAB method
  2. PCR amplification: primer sequence COI-F:5'-GGTCAACAAATCATAAAGAT-3'
  3. Electrophoresis analysis: 1.5% agarose gel, expected band 658bp

Common misjudgment avoidance

Pay special attention to distinguish it from Adoxophyes honmai:

  • The terminal process of the male genitalia of the latter is hook-shaped
  • The silvery horizontal band on the forewing is not clearly interrupted
