Technical points of the method

Chromatographic conditions:A Thermo Hypersil GOLD column (1.9μm, 2.1×100mm) was used, and the mobile phase was 5% formic acid aqueous solution (A)-acetonitrile (B) gradient elution at a flow rate of 0.3mL/min.

Mass spectrometry parameters:ESI positive ion mode, monitoring ion pairs 356.05/185.02 (quantitative ion) and 356.05/335.9 (qualitative ion), and the collision energies were set to 20.5V and 6V, respectively.


Implementation and application recommendations

  1. Different matrix samples must strictly select the corresponding pre-treatment method according to the standard. Animal tissue samples are recommended to be treated with a high-speed refrigerated centrifuge
  2. When tuning the mass spectrometer, ensure that the response intensity of the parent ion 356.05 is ≥95%, and the secondary mass spectrometry fragment ratio should meet the requirements of Appendix C
  3. The standard solution must be stored at -15℃~-20℃ away from light, and the working solution must be prepared and used immediately

Technology evolution analysis

This standard has the following major improvements compared to GB/T 20195-2006: ① The detection limit is reduced by 10 times; ② A new feed matrix treatment method is added; ③ The UPLC-MS/MS technology is used to increase throughput. Since its implementation in 2020, it has become the main basis for the supervision of florfenicol in imported and exported animal products.
