Introduction
Interpretation of the core content of the standard
| Technical module | 2002 edition | Main update points of the 2020 edition |
|---|---|---|
| Detection method | Morphological identification only | New ELISA serological test (sensitivity increased by 40%) |
| Scope of application | Live cattle quarantine | Extended to Hide quarantine and epidemiological monitoring |
| Judgment standard | Single morphological judgment | Establish a three-level judgment system (negative/suspicious/positive) |
Key technical points
1. Upgrade of morphological identification
The new standard refines the morphological characteristics of the third-stage larvae: body length 19-25mm, and the structure of the valve plate spines becomes the key to identification. Compared with the 2002 version, the description of the characteristics of the second-stage larvae with pomegranate granules is added.
2. ELISA detection specifications
Typical case: A port adopted the new standard to detect imported Angus cattle, and found 3 cases of latent infection through the S/P value 55% threshold, avoiding the introduction of the epidemic.
The detection process is strictly followed: the precise ratio of 190μL diluent + 10μL serum, and the TMB color development time is controlled at 20±1 minutes.
Recommendations for the implementation of the standard
- Laboratory configuration:It must be equipped with a stereo microscope (40× or above) and an ELISA reader (450nm filter)
- Quality control: Each batch of testing must contain 3 positive and negative controls, and PCx/NCx≥3.5 is valid
- Handling of suspicious samples: It is recommended to adopt a composite verification scheme of secondary sampling + PCR verification
Analysis of technological evolution
This standard responds to the requirements of the OIE Terrestrial Animal Health Code (2019) and aligns my country's cattle myiasis quarantine with international standards. Major breakthroughs:
- Introduction of standard operating procedures for commercial test kits
- Added pathological indicators for subcutaneous cellulitis on the back
- Cleared the differential identification characteristics of Dermatophagus gracilis/Dermatophagus sinensis
