Key technical points

1. Upgrade of morphological identification

The new standard refines the morphological characteristics of the third-stage larvae: body length 19-25mm, and the structure of the valve plate spines becomes the key to identification. Compared with the 2002 version, the description of the characteristics of the second-stage larvae with pomegranate granules is added.

2. ELISA detection specifications

Typical case: A port adopted the new standard to detect imported Angus cattle, and found 3 cases of latent infection through the S/P value 55% threshold, avoiding the introduction of the epidemic.

The detection process is strictly followed: the precise ratio of 190μL diluent + 10μL serum, and the TMB color development time is controlled at 20±1 minutes.


Recommendations for the implementation of the standard

  1. Laboratory configuration:It must be equipped with a stereo microscope (40× or above) and an ELISA reader (450nm filter)
  2. Quality control: Each batch of testing must contain 3 positive and negative controls, and PCx/NCx≥3.5 is valid
  3. Handling of suspicious samples: It is recommended to adopt a composite verification scheme of secondary sampling + PCR verification
