Introduction

1. Background and significance of the standard

GB/T33117-2016 "Identification Method for Quarantine of Wheat Basal Rot" is an important standard formulated by my country for wheat basal rot (Helgaria herpotrichioides), aiming to standardize the quarantine process and detection technology of the pathogen. This standard was proposed and coordinated by the National Technical Committee for Plant Quarantine Standardization, and the drafting units include Shandong Entry-Exit Inspection and Quarantine Bureau of the People's Republic of China, Qingdao Inspection and Quarantine Technology Development Center, etc.

2. Basic information of wheat basal rot pathogen

3. Analysis of quarantine identification methods

On-site quarantine: According to SN/T2122 and SN/T2589, focus on whether there are suspected lesions on the stems and leaf sheaths of the host plant.

Laboratory quarantine: Comprehensive judgment is made through symptom inspection, PCR initial screening test and hyphae morphology observation.

Case analysis: Isolation and culture of wheat base rot pathogen

Taking soil samples as an example, the selective PDA plate separation method was used to culture the pathogen. The results showed that at 15℃, the pathogen formed characteristic brown colonies, which were easy to distinguish from other common plant pathogens.
