High performance liquid chromatography Special attention should be paid to:

  1. The mobile phase ratio is strictly controlled at methyl tert-butyl ether: acetonitrile = 35:65 (v/v)
  2. The column temperature is maintained at 25±0.5℃. Temperature fluctuations will cause retention time shift
  3. The detection wavelength of 470nm corresponds to the maximum absorption peak of astaxanthin

Implementation suggestions

Case: An astaxanthin oil test showed a forward peak. After investigation, the following results were found:

  • Incomplete enzymatic hydrolysis: add 50μL cholesterol esterase working solution
  • Insufficient petroleum ether extraction: increase to 3 extractions
  • Nitrogen blowing temperature is too high: control below 40℃
