1. Standard Overview

This standard specifies the enzyme activity and impurity detection methods of proteinase K, which is applicable to the production and quality control of biochemical reagent products. This standard was proposed and coordinated by the China National Institute of Standardization, and the drafting units include many universities, scientific research institutions and enterprises.

2. Background of Standard Formulation

Proteinase K is an important biochemical reagent and is widely used in the field of biotechnology. With the expansion of its application scope, the requirements for product quality and detection methods are also increasing. The formulation of this standard aims to standardize the detection process and ensure the consistency and reliability of the product.

3. Detection Principle and Method

Proteinase K hydrolyzes bovine hemoglobin by catalyzing the cleavage of peptide bonds to produce phenolic amino acids. Under alkaline conditions, Folin's reagent reduces it to molybdenum blue and tungsten blue, and the absorbance change is proportional to the tyrosine content, thereby calculating the enzyme activity.
