Introduction
Analysis of the core content of the standard
This standard establishes a physiological activity evaluation system for three types of enzyme preparations: lipase, protease and carbohydrate hydrolase. It simulates the animal/human digestive environment through an in vitro digestion model and adopts the gastric-intestinal two-stage digestion method to determine the digestion promotion rate.

Sample processing specifications
Solid enzyme preparations need to be ground through a 0.25mm sieve, and the working solution concentration is uniformly 0.5mg/mL; liquid enzyme preparations are diluted to 0.5μL/mL, and the actual addition amount needs to be adjusted according to the product instructions.
Construction of digestive model
Formula of simulants:
- Food: starch + egg white + soybean oil (3:1:1)
- Feed: corn + bran + soybean meal (1:1:1)
Digestive parameter control: 37℃, 120 times/min oscillation, pH gradient control (gastric stage 2.0→intestinal stage 7.0)
Analysis of standard evolution
This standard is the first to establish an evaluation system for the physiological activity of enzyme preparations rather than simply the enzyme activity. The innovations include:
1. Introducing an in vitro digestion model to simulate the real digestive environment
2. Defining the calculation formula for digestion promotion rate: P=(W₂-W₁)/W₀×100%
3. Distinguishing the evaluation conditions of the two types of feed/food simulants
Application suggestions
- The experiment needs to be repeated ≥3 times, and the residue needs to be cleaned with acetone and dried at 105℃
- The result should include: enzyme preparation name, manufacturer, concentration, three major nutrients content and digestion promotion rate
- It is recommended to cooperate with the standard operation of gastric/intestinal simulation solution preparation in Appendix A/B
